New scientific evidence points to the ability of Affron®, Pharmactive’s standardized saffron extract (Crocus sativus L.), to help the body adapt more effectively to stress by supporting HPA axis balance.
A study published in Nutrients (Nov 2023), led by Chae-Young Kim, PhD, investigated the effects of Affron® in an in vivo stress model. The findings suggest that the extract can restore proper regulation of the hypothalamic–pituitary–adrenal (HPA) axis, improving resilience to occasional stress.
The stress signal
Key Study Insights
The trial involved 40 Wistar rats divided into four groups. Over four weeks:
-
Two groups received oral doses of Affron® (100 mg/kg and 200 mg/kg).
-
One stress-exposed group received no treatment.
-
A control group was not exposed to stress.
Results:
-
Affron® reduced hypothalamic levels of arginine vasopressin and CRF, lowering downstream stress hormone release.
-
The high-dose group showed stronger negative feedback regulation via decreased glucocorticoid receptor (GR) expression, helping prevent stress-related receptor resistance.
-
Rats maintained higher levels of Brain-Derived Neurotrophic Factor (BDNF), supporting neuronal plasticity and emotional adaptation.
-
Behavioral improvements were observed: in the Sucrose Preference Test, Affron® revived interest in sugar among stressed rats—an indicator of restored motivation and reduced anhedonia.
“Excessive activation of the HPA axis contributes to allostatic overload, which accelerates both mental and physical wear,” explains Inés Morán, PhD, Scientific Studies Manager at Pharmactive. “Affron® showed the capacity to rebalance stress pathways and support resilience.”
A Botanical with Growing Scientific Support
While saffron has a long tradition in wellness, modern science continues to uncover its role in mental balance. Previous clinical studies have demonstrated Affron®’s efficacy in reducing occasional stress, improving mood, and enhancing sleep quality across different populations, including adolescents.
“This study expands our understanding of saffron’s mechanisms,” notes Carlos Rodríguez, Communications Manager at Pharmactive. “Affron® goes beyond serotonin modulation, showing potential to normalize HPA axis activity and strengthen resilience to chronic stress.”

Conclusion: Nature and Science Against Stress
With global challenges increasing stress levels worldwide, demand for natural, science-backed solutions is rising. Affron® emerges as a promising ingredient for nutraceutical formulations that address the roots of stress while supporting overall mental and physical well-being.
